Avtofagični mehanizmi pri staranju in nevrodegeneraciji

Staranje je posledica neustavljivega in naključnega kopičenja poškodb v biomolekulah, ki so ključne za pravilno delovanje celic. Čeprav je ta proces nepredvidljiv in neobvladljiv, je nazadovanje povezano s staranjem v veliki meri določeno z genetskimi in zunanjimi dejavniki. Znano je, da številne genske mutacije in zdravljenja podaljšujejo življenjsko dobo različnih organizmov, od enoceličnih kvasovk do primatov. Vse bolj očitno postaja tudi, da večina takšnih posegov na koncu vpliva na mehanizme celičnega odziva na stres, kar kaže, da je dolgoživost tesno povezana s sposobnostjo organizma, da se učinkovito spoprijema z notranjim in zunanjim stresom. Predstavil bom naša opažanja, ki kažejo, da je pomanjkljiva odstranitev poškodovanih mitohondrijev ključni dogodek pri nevrodegeneraciji. Mitofagija je selektivna vrsta avtofagije, ki omogoča odstranjevanje poškodovanih mitohondrijev, in je glavna pot razgradnje, s katero celice uravnavajo število mitohondrijev kot odgovor na svoje presnovno stanje. Te ugotovitve kažejo, da je mitofagija lahko smiselna tarča za razvoj inovativnih in učinkovitih terapevtskih posegov za boj proti človeškim nevrodegenerativnim motnjam.

 

O predavatelju: Nektarios Tavernarakis je zaslužni profesor molekularne sistemske biologije na Medicinski fakulteti Univerze na Kreti, kjer vodi laboratorij za nevrogenetiko in staranje. Je predsednik upravnega odbora Fundacije za raziskave in tehnologijo v Heraklionu. Leta 2020 je bil izvoljen za podpredsednika Evropskega raziskovalnega sveta (ERC), leta 2022 pa za predsednika upravnega odbora in izvršnega odbora Evropskega inštituta za inovacije in tehnologijo (EIT).

Autophagic mechanisms in ageing and neurodegeneration

Ageing is driven by the inexorable and stochastic accumulation of damage in biomolecules vital for proper cellular function. Although this process is unpredictable and uncontrollable, the decline associated with ageing is broadly influenced by genetic and extrinsic factors. Numerous gene mutations and treatments have been shown to extend the lifespan of diverse organisms ranging from the unicellular yeast to primates. It is becoming increasingly apparent that most such interventions ultimately interface with cellular stress response mechanisms, suggesting that longevity is intimately related to the ability of the organism to effectively cope with both intrinsic and extrinsic stress. I will present our observations indicating that defective removal of damaged mitochondria is a pivotal event in neurodegeneration. Mitophagy is a selective type of autophagy mediating elimination of damaged mitochondria, and the major degradation pathway, by which cells regulate mitochondrial number in response to their metabolic state. These findings highlight mitophagy as a potential target for the development of innovative, effective therapeutic interventions towards battling human neurodegenerative disorders.

 

About the lecturer: Nektarios Tavernarakis is a distinguished professor of molecular systems biology at the Medical School of the University of Crete where he is heading the Neurogenetics and Ageing Laboratory. He is the chairman of the board of directors at the Foundation for Research and Technology in Heraklion. He was elected vice president of the European Research Council (ERC) in 2020, and chairman of the European Institute of Innovation and Technology (EIT) governing board and executive committee in 2022.
