Vabimo vas na 11. predavanje iz sklopa »Kolokviji na IJS« v akademskem letu 2025/26, ki bo izjemoma v ponedeljek, 4. maja 2026, ob 13:00 v Veliki predavalnici Instituta »Jožef Stefan« na Jamovi cesti 39 v Ljubljani. Predavanje bo v angleščini. Na voljo bodo kava, pijača in prigrizki.

 

 

Kako celice uporabljajo novo odkrito signalno pot za obrambo in programirano celično smrt

Vse žive celice morajo biti sposobne zaznati nevarnost, se boriti proti okužbam in se odločiti, kdaj sprožiti programirano celično smrt. Čeprav so ti procesi osrednji za delovanje imunskega sistema, ustrezni molekularni mehanizmi postajajo razumljivi šele v zadnjem času. Pomembno vlogo pri tem imajo domene TIR (receptor Toll/interleukin 1), ki se pojavljajo v beljakovinah, povezanih z imunskim odzivom, pri živalih, rastlinah in bakterijah. S kombinacijo različnih strukturnobioloških pristopov, med drugim kriogene elektronske mikroskopije, uklona elektronov na mikrokristalih in serijske femtosekundne kristalografije, smo proučili signalne komplekse, ki jih tvorijo domene TIR. Ugotovili smo, da številne med njimi delujejo kot encimi, ki cepijo nukleotide, nastale signalne produkte pa smo določili z jedrsko magnetno resonanco. Ti kompleksi tvorijo filamente, nov način signaliziranja, ki se razlikuje od klasičnih poti, pa smo poimenovali SCAF (signaliziranje s kooperativnim nastajanjem kompleksov). Ta mehanizem odpira možnosti za razvoj novih pristopov k zdravljenju vnetnih motenj in bakterijskih okužb pri ljudeh ter za zaščito rastlin pred boleznimi.

 

O predavatelju: Boštjan Kobe je profesor strukturne biologije na Univerzi v Queenslandu in eden vodilnih svetovnih raziskovalcev na področjih beljakovinske strukture, imunosti in razvoja strukturnobioloških metod. Je prejemnik številnih mednarodnih priznanj, med drugim članstva v britanski Kraljevi družbi in Avstralski akademiji znanosti ter naziva Ambasador Republike Slovenije v znanosti.

How cells use a newly identified signalling mechanism for defense and programmed cell death

All living cells must be able to detect danger, fight infection and decide when to trigger programmed cell death. Although these processes are central to immunity, many of the molecular mechanisms behind them are only now being uncovered. One important group of players are TIR (Toll/interleukin-1 receptor) domains, found across animals, plants and bacteria in proteins that mediate immune responses. Using a range of structural biology approaches, including cryogenic electron microscopy, microcrystal electron diffraction and serial femtosecond crystallography, we characterized the signalling assemblies formed by TIR domains. We found that many TIR domains act as enzymes that cleave nucleotides, and we used nuclear magnetic resonance to identify the signalling products they generate. These assemblies form filaments, and the resulting signalling mechanism, which is distinct from classical pathways, has been termed SCAF (signalling by cooperative assembly formation). This emerging mechanism may open new avenues for treating inflammatory disorders and bacterial infections, as well as for protecting plants from disease.

 

About the lecturer: Boštjan Kobe is a professor of Structural Biology at the University of Queensland and one of the world’s leading researchers in protein structure, immunity, and structural‑biology method development. He is the recipient of numerous international distinctions, including election to the Royal Society and the Australian Academy of Science, as well as the title Ambassador of the Republic of Slovenia in science.
