Vabimo vas na 1. predavanje iz sklopa »Kolokviji na IJS« v akademskem letu 2024/25, ki bo v sredo, 18. septembra 2024, ob 13:00 v Veliki predavalnici Instituta »Jožef Stefan« na Jamovi cesti 39 v Ljubljani. Predavanje bo v angleščini. Na voljo bodo kava, pijača in prigrizki.
Od izvornih celic do laboratorijsko gojenih pametnih možganov

»Mini možgane« je v laboratoriju mogoče vzgojiti iz velike zbirke tridimenzionalnih kultur matičnih celic, s katerimi poskušamo posnemati številne vidike razvoja in delovanja pravih možganov. Naš laboratorij se osredotoča na funkcionalno karakterizacijo »mini možganov«, pripravljenih iz induciranih pluripotentnih matičnih celic (iPSC) miši in ljudi, predvsem z vidika imunskega odziva in elektrofiziološke aktivnosti. Prvi del predavanja bo posvečen našim nedavnim dosežkom pri uporabi integracijske analize transkriptoma in proteoma za preučevanje in spreminjanje imunskega odziva v »mini možganih« iz sogojenih mišjih iPSC in celic mikroglije. Drugi del predavanja bo obravnaval vpliv dogodka, ki posnema možgansko kap, in virusnih okužb na aktivnost elektrofiziološke mreže v »mini možganih« iz človeških iPSC. Z navedenimi primeri tekočih raziskav želimo prispevati k razvoju in uporabi funkcionalno kompetentnih laboratorijsko gojenih človeških »mini možganov«.
O predavatelju: Peter Ponsaerts je profesor na Fakulteti za medicino in zdravstvene vede na Univerzi v Antwerpnu. Je predsednik Belgijskega združenja za celično in razvojno biologijo in je nosilec patenta, licenciranega za Argos Therapeutics in Ci-Maas.

From stem cells to smart brains in the lab

Lab grown mini-brains encompass a large collection of in vitro stem cell-derived three-dimensional culture systems that aim to recapitulate multiple aspects of in vivo brain development and function. Our laboratory focusses on the functional characterisation of murine and human induced pluripotent stem cell (iPSC)-derived mini-brains in terms of immune competence and electrophysiological activity. The first part of the lecture will discuss our recent developments in the application of transcriptome-proteome integration analysis to study and modulate detrimental immune responses in microglia-containing murine iPSC-derived mini-brains. The second part of the lecture will discuss the influence of a stroke-like event and a viral infection on electrophysiological network activity in human iPSC-derived mini-brains. Providing these examples of ongoing research, we wish to contribute to a new research era using functionally competent human lab-grown mini-brains.
About the lecturer: Peter Ponsaerts is a professor at the Faculty of Medicine and Health Sciences at the University of Antwerp. He is the president of the Belgian Society of Cell and Developmental Biology and holds a patent licenced to Argos Therapeutics and Ci-Maas.
