<p class=MsoNormal style='text-align:justify'>Strjevanje krvi je fiziološki
proces, pri katerem telo ščiti integriteto žil. Klinični preizkusi strjevanja
krvi se redno izvajajo pri ugotavljanju motenj pri strjevanju krvi, kot sta
hemofilija A in B, vendar so ti preizkusi (testi) sedaj omejeni zaradi
motenj, ki so v krvi pri nekaterih bolnikih. Treba je bilo razviti analizo
strjevanja krvi, ki je ponovljiva, zanesljiva in uporabna pri bolnikih s
sistemskim lupusom, pri katerih so navzoča protitelesa (lupusni
antikoagulanti). Na podlagi več desetletij temeljnih raziskav delovanja
kompleksa protrombinaze, ki je sestavljen iz serinskih proteaz, faktorja Xa
(FXa) in njegovega kofaktorja, faktorja Va (FVa), ob navzočnosti vodotopnega
fosftidilserina (PS), smo razvili in začeli študij zanimive nove spojine
PS, ki vsebuje nasičene alkilne verige, sestavljene iz samo šestih ogljikov
(C6PS). Z uporabo te vodotopne molekule PS nam je v določenih okoliščinah
uspelo obiti omejitve, ki jih povzročajo lupusni antikoagulanti. Za to novo
učinkovino smo že pridobili ameriški patent. Uspešen razvoj tega testa
strjevanja krvi bo zmanjšal število napačnih diagnoz motenj strjevanja krvi in
odpravil potrebo po dragih nadaljnjih testiranjih.<span style='font-size:12.0pt;

to bypass limitations of existing clotting assays <o:p></o:p></span></b></p>

<p class=MsoNormal style='text-align:justify'><span lang=EN-US>Blood clotting
is the mean by which the body protects the integrity of blood vessels. Clinical
tests, or clotting assays, are regularly performed to characterize bleeding
disorders such as hemophilia A and B but are currently limited due to
interferences present in the blood of some patients. There is a pressing need
to develop a coagulation assay that is reproducible, reliable and that can be
used for patients containing Lupus agents. Based on several decades of basic
research in the regulation of the prothrombinase complex, which consists
of a serine protease, factor Xa (FXa), and a protein cofactor, factor Va (FVa)
by water soluble Phosphatidyl Serine (PS), we have developed and started to
study an exciting novel PS compound containing saturated diacyl chains of
only six carbons (C6PS). Using this soluble PS molecule, we have been able
under ceratin circumstances to bypass the limitations caused by lupus
anticoagulants and we have already obtained an US Patent for this novel
reagent. Successful development of this assay will decrease misdiagnoses of
bleeding disorders and will remove the need for expensive subsequent testing.</span><span
lang=EN-US style='font-size:12.0pt'><o:p></o:p></span></p>
